In Vitro Study Highlights Role of Mitochondria in Regulating Cellular NAD

A new study underscores how mitochondria serve as a ‘buffer’ for NAD, ensuring that critical metabolic processes continue under stress or nutrient deprivation, according to the authors.

A new study published in Nature Metabolism highlighted the role of mitochondria as a regulator of cellular nicotinamide adenine nucleotide (NAD) levels.

The new research demonstrates that while cells can adapt to NAD decline caused by aging, disruptions in mitochondrial NAD levels have a more severe impact, potentially contributing to metabolic imbalances and age-related conditions such as neurodegenerative diseases.

Mitochondria, the authors found, serves as a “buffer” to maintain NAD levels across cellular compartments, ensuring that critical metabolic processes continue under stress or nutrient deprivation. The researchers also identified key enzymes and transporters involved in regulating NAD availability, potentially opening new avenues for interventions targeting mitochondrial health.

“The main takeaway is that NAD decline is generally well tolerated by cells unless it originates in mitochondria,” the authors wrote. “Mitochondria can act as a cellular buffer for NAD, and their dysfunction may exacerbate neurodegenerative diseases known to involve mitochondrial deficiencies,” said Mathias Zeigler, study author and professor of biomedicine at the University of Bergen in Norway.
